Potential role of tocilizumab in severe gastrointestinal barrier damage after CAR T-cell therapy

چکیده
We report a septicemia and disseminated candidiasis due to delayed gastrointestinal mucosae repair in patient treated with tocilizumab after anti-CD19 CAR T-cell therapy. Tocilizumab could have inhibited intestinal tissue furthered bacteria translocation leading the invasion of mucosa by yeasts as IL-6 is known be involved mucosal wound healing.

CAR T-cell Therapy of Hematologic Malignancies: An Update in Targeted Antigens
Immunotherapy with genetically engineered T-cells that express the chimeric antigen receptor (CAR) has raised hopes for the treatment of pediatric malignancies. Although CAR T-cell development is on a fast-moving pace and evolution, the context of exploring novel targetable antigens has been neglected. In this review study, we analyze the prominent hematologic antigens targeted by engineered T-...

Chimeric antigen receptor (CAR)-engineered T cells (CAR-T cells) have yielded unprecedented efficacy in B cell malignancies, most remarkably in anti-CD19 CAR-T cells for B cell acute lymphoblastic leukemia (B-ALL) with up to a 90% complete remission rate. However, tumor antigen escape has emerged as a main challenge for the long-term disease control of this promising immunotherapy in B cell mal...
متن کاملNext frontiers in CAR T-cell therapy
Chimeric antigen receptor (CAR) T-cell therapy is entering a new era, transitioning from an experimental approach being tested in a handful of centers to a more mainstream and broadly investigated therapeutic platform with significant efforts directed towards commercial translation. CARs are synthetic receptors engineered and transduced into T cells to redirect T-cell cytotoxicity by recognitio...
متن کاملGlobal Manufacturing of CAR T Cell Therapy
Immunotherapy using chimeric antigen receptor-modified T cells has demonstrated high response rates in patients with B cell malignancies, and chimeric antigen receptor T cell therapy is now being investigated in several hematologic and solid tumor types. Chimeric antigen receptor T cells are generated by removing T cells from a patient's blood and engineering the cells to express the chimeric a...
متن کاملCAR-T cell therapy in gastrointestinal tumors and hepatic carcinoma: From bench to bedside
The chimeric antigen receptor (CAR) is a genetically engineered receptor that combines a scFv domain, which specifically recognizes the tumor-specific antigen, with T cell activation domains. CAR-T cell therapies have demonstrated tremendous efficacy against hematologic malignancies in many clinical trials. Recent studies have extended these efforts to the treatment of solid tumors. However, th...
